    Overview

    Nap-22 (also known as BASP1) is a neuron-enriched, membrane-associated protein implicated in regulating axonal growth, neurite outgrowth, and synaptic function through interactions with phospholipids and signaling complexes at the plasma membrane. It participates in activity-dependent neuronal plasticity and has been linked to cytoskeletal organization and vesicle trafficking processes that shape growth cone dynamics. Nap-22 has also been studied in the context of transcriptional regulation via nuclear interactions, connecting extracellular cues to gene expression programs relevant to neuronal differentiation. Altered Nap-22 expression or localization has been associated with neurodevelopmental and neurodegenerative research areas where membrane signaling and synaptic remodeling are disrupted.

    Nap-22 CRISPR/Cas9 KO Plasmid (r) is a pool of plasmids designed for targeted disruption of the gene in rat cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.

    The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Nap-22 protein expression.

    This CRISPR knockout system enables efficient generation of -deficient cell models for investigation of Nap-22 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
