NANOS3 encodes the RNA-binding protein Nanos3, a conserved post-transcriptional regulator required for human primordial germ cell survival and maintenance. Nanos3 functions in germline specification programs by repressing translation and promoting selective mRNA turnover, integrating with pathways that control cell fate decisions, proliferation, and apoptosis during early development. Dysregulated NANOS3 activity has been associated with impaired germ cell development and infertility-related phenotypes, and altered expression has been reported in select germ cell–derived tumors. As a germline determinant, NANOS3 is a useful node for dissecting RNA regulatory networks and lineage-restricted transcriptional programs.
Nanos3 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the NANOS3 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the NANOS3 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the NANOS3 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Nanos3 protein expression.
This CRISPR knockout system enables efficient generation of NANOS3-deficient cell models for investigation of Nanos3 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
