Na+/K+-ATPase α3 Antibody (H-4) is a mouse monoclonal IgG1 kappa light chain antibody that detects Na+/K+-ATPase α3 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Na+/K+-ATPase α3 (H-4) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Na+/K+-ATPase α3 is a vital component of the sodium/potassium-ATPase complex, which is crucial for maintaining cellular homeostasis. Na+/K+-ATPase α3 is primarily located in the plasma membrane of cells, where Na+/K+-ATPase α3 functions to transport sodium and potassium ions across the membrane, thereby regulating cellular volume, membrane potential, and nutrient uptake. Proper localization of Na+/K+-ATPase α3 is essential for ion transport, as any disruption can lead to cellular dysfunction and contribute to various pathophysiological conditions. The Na+/K+-ATPase complex is composed of multiple isoforms of three subunits: α, β, and γ, with the α subunit being responsible for ATP binding and cation transport. The β subunit plays a critical role in the correct folding and membrane insertion of the α subunit, while the γ subunit enhances the affinity of the complex for ATP in specific nephron segments. Understanding the localization and function of Na+/K+-ATPase α3 is important for elucidating Na+/K+-ATPase α3′s role in cellular physiology and potential implications in disease states.

What application is the blocking peptide sc-365744 P appropriate for?

Asked by: jenniferc
Thank you for your question. The blocking peptide is intended for use as a negative control, by pre-adsorbing the mouse monoclonal antibody against the antigen. For full protocol details, please contact our Technical Services Department or view our online protocol here: https://www.scbt.com/scbt/resources/protocols/peptide-neutralization

Rated 2 out of 5 by from Weak BandsI tried different concentrations of antibody in WB experiments. However, the bands were too weak and difficult to quantify.

Rated 5 out of 5 by from Works well on blotsWe compared the H-4 Na,K-ATPase alpha3 antibody with the old XVI-F9G10 monoclonal, side by side. We diluted the F9G10 ascites 1:750,000 as we usually do. We diluted the H-4 antibody 1:500 (more dilute than the 1:200 recommended). The blot was of a crude homogenate of mouse brain, and both blots were almost clean of background bands. Although we did not yet test it on tissue that has alpha1 or 2 and not alpha3, brain has all three isoforms, and we did not detect anything suspicious of crossreactivity. We have not yet tested it on tissue.
