N-Ras Antibody (F155) is a mouse monoclonal IgG1 kappa light chain antibody, specifically raised against recombinant human N-Ras p21, and demonstrates reactivity with N-Ras proteins from mouse, rat, and human origins in applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). The N-Ras protein is a small GTPase belonging to the Ras family, playing a crucial role in cellular signal transduction pathways that regulate cell proliferation, differentiation, and survival. N-Ras functions as a molecular switch by cycling between an active GTP-bound state and an inactive GDP-bound state, a process regulated by guanine nucleotide exchange factors (GEFs) and GTPase-activating proteins (GAPs). Post-translational modifications of N-Ras, such as farnesylation and palmitoylation at its C-terminal CAAX motif, are essential for proper localization to the plasma membrane and interaction with downstream effectors. These lipid modifications enable N-Ras to anchor to cellular membranes, which is critical for transmitting signals that influence cell growth and differentiation. Disruptions in these post-translational modifications can lead to mislocalization and aberrant signaling of N-Ras, contributing to oncogenesis and the progression of various cancers. N-Ras (F155) antibody is a valuable tool for researchers studying the expression, localization, and function of N-Ras in both normal physiology and disease states, aiding in the advancement of understanding cellular signaling mechanisms and the development of targeted therapies.

Hi, can this N-Ras antibody(F155) be used to detect the Nras encoded by mutated NRAS, such as NrasQ61R?

Thank you for your question. This antibody is raised against recombinant N-Ras p21 of human origin, recommended for detection of N-Ras p21 of mouse, rat and human origin by WB, IP, IF and IHC(P), we didn't test it on mutated NRAS.

Hello, I am interested in "N-Ras Antibody (F155): sc-31" for Western blot use. What were the duration, buffers, Ab concentrations, and temperature/conditions (for blocking/probing) used to QC this Ab? Thank you in advance

Thank you for your question. If you are using ECL for detection, we recommend blocking (PVDF membrane) in 5% nonfat milk + TBST for 1 hour at room temperature. We recommend using the same buffer (freshly prepared) as a diluent for both primary and secondary antibodies. If you are using Near-Infrared (NIR) Detection methods, then we recommend using our UltraCruz Blocking Reagent: sc-516214 instead of milk or BSA to reduce background. Recommended starting dilution for Western blot for the primary antibody is 1:200 (range 1:100-1:1,000). We recommend incubating primary antibody for 1 hour at room temperature, or overnight at 4C. Can I use this mouse monoclonal antibody for immunohistochemical staining of mouse tissue?

No. Since no direct conjugates are available for this antibody, we would not recommend using it for IHC in mouse tissue. See pan Ras (C-4): sc-166691 for N-Ras antibody conjugates, including AC, HRP, FITC, PE, Alexa Fluor® 488 and Alexa Fluor® 647.

SIgnals were detected from cells transfected with the NRAS expression plasmid, although their intensity was low. No signals were observed in the non-transfected cell lines; therefore, detection of endogenous NRAS may be difficult.
