N-cadherin Antibody (D-4) is a mouse monoclonal IgG1 kappa light chain antibody that detects N-cadherin of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). N-cadherin monoclonal antibody (D-4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. N-cadherin, also known as CD325 or CDH2, plays a crucial role in cell-cell adhesion, essential for maintaining tissue integrity and facilitating morphogenetic processes during development. N-cadherin is primarily located at the cell membrane, forming adherens junctions that connect adjacent cells, thereby contributing to tissue structural stability. Such localization allows N-cadherin to interact with the cytoskeleton and other signaling molecules, influencing cellular behavior and communication. The extracellular domain of N-cadherin contains five homologous repeats mediating homophilic binding, while the intracellular domain interacts with cytoplasmic proteins like β-catenin, linking to the actin cytoskeleton and regulating cell motility and tissue architecture. Understanding N-cadherin function and localization helps elucidate involvement in various physiological processes and implications in diseases like cancer, where altered expression can lead to increased cell migration and metastasis.

Can N-cadherin Antibody (D-4): sc-8424 be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Thank you for your question. Yes, N-cadherin Antibody (D-4): sc-8424 has been validated for use in IHC with paraffin-embedded sections. Rated 1 out of 5 by from Antibody does not work for IHC-PI purchased this antibody based on very nice images on the website showing specifically localization of NCAD in IHC-P. I tested the antibody using multiple techniques and tissue types with no staining. I reported the problems to the company and they kindly provided a replacement vial. Unfortunately the replacement did not work either again using multiple techniques and tissue types. While this antibody may have previously worked well in IHC-P, the current product being provided does not work for IHC-P.
