Myo1a encodes myosin Ia, an actin-based motor enriched in polarized epithelial cells where it supports microvillar structure and membrane–cytoskeleton coupling at the apical surface. Myosin Ia contributes to actin filament dynamics, membrane tension, and vesicle trafficking processes that help maintain brush border organization and epithelial barrier function. Through interactions with the cortical actin network, it influences pathways governing cell polarity, absorption, and mechanical responses in intestinal epithelium. Altered MYO1A function or expression has been associated with epithelial dysregulation and has been investigated in gastrointestinal pathobiology, including contexts of barrier impairment and tumorigenesis.
Myosin Ia CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Myo1a gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Myo1a together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Myo1a open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Myosin Ia protein expression.
This CRISPR knockout system enables efficient generation of Myo1a-deficient cell models for investigation of Myosin Ia signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
