Munc13-1/2/3 Antibody (32) is a mouse monoclonal IgG1 kappa light chain antibody that detects Munc13-1/2/3 protein of mouse and rat origin by western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). Munc13-1/2/3 (32) antibody is available as the non-conjugated form. Munc13 proteins, which include Munc13-1, Munc13-2, and Munc13-3, are critical components of synaptic transmission, primarily expressed in neurons, with Munc13-2 also having a splice variant that is ubiquitously expressed. These proteins play a vital role in the regulation of neurotransmitter release by binding to Syntaxin, a key player in the exocytosis process. Munc13-1, in particular, is essential for synaptic vesicle maturation and is involved in the central priming function necessary for the exocytosis of synaptic vesicles at glutamatergic synapses. The presence of phorbol ester binding C1- and C2-domains within Munc13 proteins allows them to interact with calcium, phospholipids, and diacylglycerol, which are crucial for their regulatory functions. Munc13-1 is predominantly localized in presynaptic terminals, where Munc13-1 interacts with Doc2α and msec7-1, facilitating the co-localization of these proteins at the active zone, a specialized area of the presynaptic membrane characterized by high membrane turnover. Munc13-2 is expressed in rostral brain regions, while Munc13-3 is primarily found in the cerebellum, highlighting the diverse roles these proteins play in neuronal function and signaling.
