MTHFD1 Antibody (A-8) is a mouse monoclonal IgG1 kappa light chain antibody that detects MTHFD1 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). MTHFD1 (A-8) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Methylenetetrahydrofolate dehydrogenase 1 (MTHFD1) is a crucial 935-amino acid, folate-dependent enzyme that plays a vital role in the interconversion of tetrahydrofolate derivatives, which are essential for the synthesis of purines, methionine, and thymidylate. MTHFD1 is located in the cytosol and exists as a homodimer composed of three distinct subunits: 5,10-methylenetetrahydrofolate dehydrogenase, 5,10-methenyltetrahydrofolate cyclohydrolase, and 10-formyltetrahydrofolate synthetase, each contributing unique enzymatic activities. MTHFD1′s structural organization, with its N-terminal domain responsible for dehydrogenase and cyclohydrolase activities and a larger C-terminal synthetase domain, is critical for its function in folate metabolism. Mutations in MTHFD1 gene in pregnant women have been linked to an increased risk of neural tube defects in offspring, highlighting MTHFD1′s significance in developmental biology. MTHFD1 is also implicated in migraine pathophysiology, particularly in cases of migraine with aura, due to MTHFD1′s role in folate metabolism.

I am using MTHFD1 (A-8): sc-271412 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?
