MTHFD1 Antibody (A-1) is a mouse monoclonal IgG2a antibody that detects methylenetetrahydrofolate dehydrogenase 1 (MTHFD1) in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). MTHFD1 is a crucial 935-amino acid, folate-dependent enzyme that facilitates the interconversion of tetrahydrofolate derivatives, which are essential for the synthesis of purines, methionine, and thymidylate, thereby playing a vital role in cellular metabolism and DNA synthesis. MTHFD1 is located in the cytosol and exists as a homodimer composed of three distinct subunits: 5,10-methylenetetrahydrofolate dehydrogenase, 5,10-methenyltetrahydrofolate cyclohydrolase, and 10-formyltetrahydrofolate synthetase, each contributing unique enzymatic activities. The structural organization of MTHFD1, with its N-terminal domain responsible for dehydrogenase and cyclohydrolase activities and a larger C-terminal synthetase domain, is critical for its function, as this allows for efficient catalysis and regulation of folate metabolism. Mutations in the MTHFD1 gene have been linked to an increased risk of neural tube defects in offspring of pregnant women, highlighting MTHFD1′s significance in developmental biology and maternal health. MTHFD1′s involvement in folate metabolism is associated with migraine pathophysiology, particularly in cases of migraine with aura, underscoring MTHFD1′s relevance in both metabolic and neurological contexts.
