MSH2 Antibody (D-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects MSH2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). MSH2 (D-6) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. MSH2 plays a critical role in the DNA mismatch repair (MMR) system, which is essential for maintaining genomic stability by correcting errors that occur during DNA replication. Deficiencies in this repair mechanism can lead to microsatellite instability, a hallmark of several cancers, including hereditary nonpolyposis colorectal cancer (HNPCC). MSH2′s importance extends beyond HNPCC, as studies have shown that 10 to 45% of various cancers, such as pancreatic, gastric, breast, ovarian, and small cell lung cancers, also exhibit microsatellite instability, indicating that MMR dysfunction may be a common feature in tumor initiation and progression. Additionally, the identification of additional homologs, PMS1 and PMS2, which are also implicated in MMR and mutated in HNPCC patients, underscores MSH2′s significance in cancer biology and highlights MSH2′s potential as a target for therapeutic intervention and diagnostic applications.

I know D-6 is the replacement of N-20. Could you tell me does they have the same immunogen amino acid sequences? Their applications look like the same.

Thank you for your question. The two antibodies have overlapping immunogen sequences, but they are not the same. This new mouse monoclonal antibody, MSH2 (D-6): sc-376384, is raised against a recombinant protein corresponding to amino acids 1-300 of MSH2 of human origin. The rabbit polyclonal antibody, MSH2 (N-20): sc-494, was raised against a short peptide sequence mapping at the N-terminus of MSH2 of human origin. We do offer a different monoclonal antibody, MSH2 (E-7): sc-365052, which is raised against the same peptide used to generate the rabbit polyclonal (N-20) antibody.

What is the recommended fixation method for immunofluorescence staining with MSH2 (D-6): sc-376384 monoclonal antibody?

It gave a strong signal when used at 1:500 dilution in 5% milk+ TBS-tween and developed with mouse-IgGκ BP-HRP(SCBT) at 1:1000 dilution in 5% milk+ TBS-tween.

Rated 5 out of 5 by from Highly potent and very specific antibodyThis monoclonal antibody was independently tested in-house by Kinexus Bioinformatics Corporation and validated for use for Western blotting. There was very strong detection of the ~100 kDa target protein in an A431/HepG2/MCF7 cell lysate mix, and weak detection in mouse tissues (brain/testes/thymus/muscle mix). No major cross-reactivities were observed at 2 µg/ml antibody concentration with off-target proteins in the mouse tissues tested or in the A431/HepG2/MCF7 cell lysate mix.
