MLH1 Antibody (D-2) is a mouse monoclonal IgG1 antibody that detects MLH1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). MLH1 plays a crucial role in the DNA mismatch repair (MMR) pathway, which is vital for maintaining genetic stability by correcting errors that occur during DNA replication. MLH1 (D-2) antibody enables detection of deficiencies that can lead to microsatellite instability and an increased risk of various cancers, including hereditary nonpolyposis colorectal cancer (HNPCC). MLH1 is predominantly located in the nucleus, where MLH1 forms a complex with other MMR proteins to identify and repair mismatched DNA bases. This localization allows for immediate response to DNA damage, thereby preventing the accumulation of mutations that could lead to tumorigenesis. Loss of MLH1 expression is often associated with a mutated phenotype in tumors, highlighting MLH1′s importance as a biomarker for cancer diagnosis and prognosis. Anti-MLH1 antibody (D-2) is particularly valuable for researchers studying the implications of MMR deficiencies in cancer biology and therapeutic responses.

When should the TransCruz reagent sc-166625 X be used instead of the standard form sc-166625?

    Asked by: DefinitelyNotMatt
Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.
