    SEE ALSO...

MKP-6 Antibody (4B5-E6) is a mouse monoclonal IgG2a kappa light chain antibody that detects MKP-6 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-MKP-6 antibody (4B5-E6) is available as the non-conjugated format. MAP kinase phosphatase 6 (MKP-6), also known as dual-specificity phosphatase 14 (DUSP14), is a 198 amino acid protein that plays a crucial role in cellular signaling by dephosphorylating key mitogen-activated protein kinases (MAPKs) such as ERK, JNK, and p38, thereby acting as a negative regulator of CD28 signaling in T cells. This regulation is particularly important for maintaining immune homeostasis and preventing overactivation of T cells, which can lead to autoimmune diseases. MKP-6 is characterized by a conserved catalytic core typical of the MKP family, but also possesses unique N- and C-terminal regions that distinguish MKP-6 from other members of the family. Notably, MKP-6 interacts specifically with the cytoplasmic tail of CD28 at Tyr 200, a critical interaction for T cell activation, although this residue can be mutated to Phe 200 without affecting binding. MKP-6 is expressed ubiquitously across various tissues, with higher expression levels observed in certain cell types, underscoring its significance in diverse biological processes.

I am using MKP-6 (4B5-E6): sc-517023 for immunofluorescence, what dilution of this antibody should I use?

    Asked by: Trav11
Thank you for your question. We recommend a starting dilution of 1:50. However, optimal antibody concentration should be determined by titration. Please find our full immunofluorescence protocol here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining
