Ccl20 encodes the chemokine macrophage inflammatory protein-3 alpha (MIP-3α), a secreted ligand for CCR6 that directs chemotaxis of immature dendritic cells, memory T cells, and B cells to sites of epithelial activation. Induced by NF-κB and proinflammatory cytokines, CCL20 participates in leukocyte trafficking, mucosal immune surveillance, and organization of inflammatory microenvironments. Dysregulated CCL20–CCR6 signaling has been associated with chronic inflammatory processes and autoimmune pathobiology, and it is frequently studied in the context of epithelial barrier dysfunction and tumor-associated immune cell recruitment. In mice, Ccl20 provides a tractable node for dissecting chemokine-driven cell migration and cytokine networks in immunology and inflammation research.
MIP-3α CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Ccl20 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Ccl20 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Ccl20 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish MIP-3α protein expression.
This CRISPR knockout system enables efficient generation of Ccl20-deficient cell models for investigation of MIP-3α signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
