Mel-CAM Antibody (2Q401) is a mouse monoclonal IgG1 antibody that detects Mel-CAM in mouse, rat, human, canine, and rabbit samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and flow cytometry (FCM). Mel-CAM, also known as MCAM, MUC18, or CD146, is a transmembrane glycoprotein encoded by the human Mel-CAM gene, which plays a crucial role in cell adhesion and is a member of the immunoglobulin superfamily. This protein consists of 603 amino acids, featuring a signal peptide, five immunoglobulin-like domains, a transmembrane region, and a short cytoplasmic tail, which are essential for its function. Mel-CAM is predominantly expressed in advanced primary and metastatic melanomas, with its presence correlating strongly with the progression of metastatic disease, as Mel-CAM is found in 80% of advanced primary human melanomas but absent in normal melanocytes. Mel-CAM′s importance lies in its ability to initiate an outside-in signaling pathway involving protein tyrosine kinases such as Fyn, FAK, and paxillin, which subsequently influences the dynamics of actin cytoskeleton rearrangement. This function is vital not only for tumor progression but also for maintaining thymic architecture and function, highlighting Mel-CAM′s significance in both cancer biology and immune system integrity. Anti-Mel-CAM antibody (2Q401) is an invaluable tool for researchers investigating the mechanisms of melanoma progression and the role of cell adhesion in tumor biology.
