Mouse Ccl8 encodes monocyte chemoattractant protein-2 (MCP-2), a CC chemokine that coordinates leukocyte trafficking by signaling through chemokine receptors such as CCR1, CCR2, and CCR5. MCP-2 shapes inflammatory chemotaxis, regulates myeloid cell recruitment, and contributes to crosstalk between innate and adaptive immune compartments within damaged or infected tissues. Ccl8 expression is commonly induced downstream of NF-κB and cytokine-driven programs, linking pattern-recognition and inflammatory signaling to cell migration. Dysregulated MCP-2 activity has been implicated in chronic inflammatory states, autoimmune phenotypes, and tumor-associated immune infiltration, making it relevant for mechanistic studies of immune microenvironments.
MCP-2 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Ccl8 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Ccl8 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Ccl8 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish MCP-2 protein expression.
