MAD2 Antibody (C-10) is a mouse monoclonal IgG2b kappa light chain antibody that detects MAD2 of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). MAD2 antibody (C-10) is available in non-conjugated form and conjugated to agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® dyes. MAD2 plays a crucial role in mitotic spindle assembly checkpoint, ensuring accurate chromosome segregation during cell division. Through binding to unattached kinetochores during prometaphase, MAD2 prevents cell progression to anaphase until all chromosomes align and attach to the spindle apparatus. Cells lacking functional MAD2 can bypass this checkpoint, leading to aneuploidy and contributing to tumorigenesis. Research shows breast carcinoma cell lines with reduced MAD2 expression, such as T47D, complete mitosis even with nocodazole present, highlighting MAD2′s importance in genomic stability. MAD2 also associates specifically with insulin receptors but not insulin-like growth factor receptors, suggesting a unique role in insulin-related cellular responses. MAD2B, a MAD2 homolog, helps execute mitotic checkpoint control by delaying anaphase onset when kinetochore-spindle attachment remains incomplete, emphasizing MAD2 family′s significance in cell cycle regulation.
