Laminin α-4 Antibody (6C3) is a mouse monoclonal IgG1 kappa light chain antibody that detects Laminin α-4 protein of human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). Anti-Laminin α-4 antibody (6C3) is available as a non-conjugated format. Laminin α-4 is a crucial component of the extracellular matrix, specifically within basement membranes, which play a vital role in tissue architecture and cellular functions. Laminin α-4 exists as a heterotrimer composed of α, β, and γ chain subunits, and proper assembly and function are essential for maintaining the structural integrity of tissues. Laminin α-4 interacts with various cell surface receptors, including integrins and dystroglycan, facilitating cell adhesion, migration, and signaling. This interaction is particularly important in processes such as wound healing and tissue repair, where the extracellular matrix provides not only structural support but also biochemical signals that influence cell behavior. The human Laminin α-4 gene is located on chromosome 6q21 and is expressed in several tissues, including the heart, lung, ovary, and intestines, highlighting its significance in both normal physiology and potential pathological conditions.

For Western Blot, is it recommended to use denatured or non-denatured conditions with Laminin α-4 (6C3): sc-130541 antibody?

Rated 4 out of 5 by from Staining of human RCC paraffin embedded tissueThe antibody works well on human paraffin sections of renal cell carcinoma. Matching normal kidney tissue of the same RCC patient shows some Lama4 staining. Protocol After deparaffinization with xylene and ethanol block endogenous peroxidase with 0.3% hydrogen peroxidase/PBS 15 min at RT. Antigen retrieval with 10mM citrate buffer pH 6.0, block 5% BSA/PBS 1h at RT, anti-lama4 Ab dilution 1:30 in 0.5% BSA/PBS O.N. at 4C, 2nd Ab goat anti-mouse biotin (Dako, ref E0433) diution 1:500 in 0.5% BSA/PBS 45 min at RT. Strep-HRP (Dako, ref P0397) 1:200 in 0.5% BSA/PBS 30 min at RT. Develop with DAB, counterstain with Mayer's hematoxylin.
