Laminin α-4 Antibody (6C3) is a high quality monoclonal Laminin alpha-4 antibody (also designated Laminin α-4 antibody) suitable for the detection of the Laminin alpha-4 protein of human origin. Laminin α-4 Antibody (6C3) is available as the non-conjugated anti-Laminin alpha-4 antibody. Laminins are essential and abundant structural non-collagenous glycoproteins localizing to basement membranes. Basement membranes (cell-associated extracellular matrices (ECMs)) are polymers of laminins with stabilizing type IV collagen networks, nidogen, and several proteoglycans. Basement membranes are found under epithelial layers, around the endothelium of blood vessels, and surrounding muscle, peripheral nerve, and fat cells. Formation of basement membranes influences cell proliferation, phenotype, migration, gene expression, and tissue architecture. Each laminin is a heterotrimer of a, b, and g chain subunits that undergoes cell-secretion and incorporation into the ECM. Laminins can self-assemble, bind to other matrix macromolecules, and have unique and shared cell interactions mediated by integrins, dystroglycan, and cognate laminin receptors. The human Laminin a-4 gene maps to chromosome 6q21 and is expressed in adult heart, lung, ovary, small and large intestines, liver, and placenta.

For Western Blot, is it recommended to use denatured or non-denatured conditions with Laminin α-4 (6C3): sc-130541 antibody?

Did not work for western blotUsed a 1:300 dilution for WB with Sum159, Caco-2, and 293T cell lysates. No band observed.

Staining of human RCC paraffin embedded tissueThe antibody works well on human paraffin sections of renal cell carcinoma. Matching normal kidney tissue of the same RCC patient shows some Lama4 staining.
After deparaffinization with xylene and ethanol block endogenous peroxidase with 0.3% hydrogen peroxidase/PBS 15 min at RT. Antigen retrieval with 10mM citrate buffer pH 6.0, block 5% BSA/PBS 1h at RT, anti-lama4 Ab dilution 1:30 in 0.5% BSA/PBS O.N. at 4C, 2nd Ab goat anti-mouse biotin (Dako, ref E0433) diution 1:500 in 0.5% BSA/PBS 45 min at RT. Strep-HRP (Dako, ref P0397) 1:200 in 0.5% BSA/PBS 30 min at RT. Develop with DAB, counterstain with Mayer's hematoxylin.
