KLHL9/13 Antibody (D-4) is a mouse monoclonal IgG2a kappa light chain antibody that detects KLHL9/13 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). KLHL9/13 (D-4) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. KLHL9, consisting of 617 amino acids, features a BACK (BTB/kelch associated) domain, six kelch repeats, and a BTB/POZ domain, while KLHL13, also known as BKLHD2, is a 604 amino acid protein with similar structural characteristics. These proteins serve as substrate-specific adapters for an E3 ubiquitin-protein ligase complex with CUL-3 in the ubiquitin-proteasome system. Such interaction enables E3 ligases to transfer ubiquitin from E2 ubiquitin-conjugating enzymes to target proteins, marking them for degradation. KLHL9 and KLHL13′s ability to modulate protein ubiquitination maintains cellular homeostasis and regulates various biological processes, including cell cycle progression and stress response.

What is the recommended fixation method for immunofluorescence staining with KLHL9/13 (D-4): sc-166486 monoclonal antibody?

Asked by: jerojero
Thank you for your inquiry. We recommend fixing cells for 5 minutes in -10°C methanol and allowing to air dry. The full protocol can be viewed here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining

Rated 5 out of 5 by from Great antibody for WBVery good antibody for WB. You can usually see a doublet, which is both KLHL9 and KLHL13 being recognized (they are highly related).

Rated 4 out of 5 by from Great Andibody for WB and IFWork excellent in western blotting at a 1:500 dillution in non transfected and KLHL9-FLAG/mCherry transfected 293T whole cell lysates. This antibody gave me also great IF results.

Rated 5 out of 5 by from Works well in WBNicely detects band at predicted (~60 kDa) MW in 293T cell lysate. Band is most likely specific because this antibody detects KLHL9/13 in IPs of a known binding partner. If you resolve band well, it looks like a doublet, which may indicate KLHL9 at the lower MW and KLHL13 at the higher MW.
