CEP295 (KIAA1731) encodes a centrosomal protein essential for centriole assembly and centrosome maturation, supporting the formation of the pericentriolar material and proper microtubule-organizing center function. It participates in processes that coordinate centriole-to-centrosome conversion, centrosome duplication, and faithful spindle formation during mitosis, thereby contributing to genome stability. Disruption of CEP295-dependent centrosome integrity can lead to aberrant mitotic progression and chromosomal instability, cellular states frequently studied in cancer biology. CEP295 is also relevant to investigations of ciliogenesis and cell-cycle control, where centrosome defects can alter signaling dynamics and differentiation programs.
KIAA1731 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the CEP295 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the CEP295 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the CEP295 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish KIAA1731 protein expression.
