JMJD2A Antibody (D-9) is a mouse monoclonal IgG1 kappa light chain antibody that detects JMJD2A of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-JMJD2A antibody (D-9) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. JMJD2A, also known as jumonji domain containing 2A, is a 1,064 amino acid protein encoded by the human gene JMJD2A and is a member of the JHDM3 histone demethylase family. JMJD2A features a complex structure that includes one JmjC domain, one JmjN domain, two PHD-type zinc fingers, and two Tudor domains, which are crucial for enzymatic activity. JMJD2A functions primarily as a histone demethylase, specifically targeting the demethylation of tri-methylated lysine residues at positions 9 and 36 of histone H3, thereby playing a pivotal role in regulating the histone code. This selective demethylation is essential for various cellular processes, including transcriptional regulation, as JMJD2A influences chromatin structure and gene expression. JMJD2A does not demethylate histone H3 at lysine 4 or lysine 27, nor histone H4 at lysine 20, which underscores specificity and importance in epigenetic regulation. The demethylation process generates formaldehyde and succinate, further linking JMJD2A to metabolic pathways. JMJD2A is involved in transcriptional repression of ASCL2 and E2F-responsive promoters through recruitment of histone deacetylases and nuclear receptor corepressor 1 (NCOR1), highlighting a role in gene silencing. As a ubiquitously expressed nuclear protein, JMJD2A is integral to maintaining proper gene expression and cellular function across various tissues.
