ISX Antibody (E-2) is a mouse monoclonal IgG1 antibody that detects ISX protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). ISX (E-2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. ISX protein, also known as intestine-specific homeobox or Pix-1, is a 245 amino acid transcription factor that plays a crucial role in regulating intestinal gene expression and is particularly important for maintaining intestinal epithelium integrity. ISX protein contains a homeobox DNA-binding domain, a conserved 60 amino acid motif that enables DNA binding through a helix-turn-helix structure, thereby influencing transcriptional regulation. ISX protein is predominantly localized in the nucleus and is expressed at high levels in intestinal epithelial cells, where ISX may also be involved in vitamin A metabolism by regulating beta-carotene monooxygenase 1 (BCMO1) expression. ISX gene is located on human chromosome 22, which houses over 500 genes and is the second smallest human chromosome. Mutations in several genes on chromosome 22 have been linked to various developmental disorders, including Phelan-McDermid syndrome, neurofibromatosis type 2, autism, and schizophrenia, highlighting ISX significance in both normal physiology and disease.
