IRF-3 Antibody (SL-12) is a mouse monoclonal IgG1 kappa light chain antibody designed to target the Interferon Regulatory Factor 3 (IRF-3) protein with exceptional precision. Engineered against a recombinant IRF-3 fusion protein corresponding to human IRF-3 (amino acids 56-427), anti-IRF-3 antibody (SL-12) reliably detects IRF-3 from mouse, rat, and human species across a variety of applications, including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). IRF-3, also referred to as IIAE7, is a critical transcription factor in the innate immune response, playing a vital role in the detection of viral infections and the subsequent activation of type I interferons, such as IFN-β. Upon viral infection, IRF-3 undergoes phosphorylation, dimerizes, and translocates to the nucleus where IRF-3 binds to specific DNA elements to initiate the transcription of interferon genes, thereby orchestrating a robust antiviral defense mechanism. Understanding and analyzing IRF-3′s function is essential for elucidating the molecular pathways involved in immune regulation and the body′s response to pathogens. IRF-3 (SL-12) monoclonal antibody is available in both non-conjugated form and a variety of conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing versatile tools for researchers to investigate the intricate roles of IRF-3 in cellular processes and disease states.

I want to perform ChIP-seq of IRF3 as done by the ENCODE team; they used your antibody sc-9082 for all their experiments. Sadly you discontinued it, and your new IRF3 AB is not validated for ChIP. Can you help me get the appropriate product from you?

Asked by: Arianna
Thank you for contacting Santa Cruz Biotechnology . For ChIP we recommend using sc-33641 X, 200 µg/0.1 ml. https://www.scbt.com/resources/protocols/chromatin-immunoprecipitation-assays

Is this antibody able to detect IRF3 dimers ?

Asked by: LeeL
Thank you for your question. We have not detected IRF-3 dimers with this antibody, however, it's possible that the antibody could detect a dimer if it was present in the sample.

You report that sc-33641 detects IRF3 of mouse and human origin, yet all your examples are human. Do you have an example showing positive WB in mouse cells/tissue, with verification it is NOT present in a KO or is induced in a TG?

Asked by: jjmaher
Thank you for your question. We do not have data in mouse cells or tissues at this time. The antibody is recommended for detection of IRF-3 of mouse, rat and human origin based on sequence identity. If the antibody does not work in any of the recommended applications or species, please contact our Technical Service Department and we would be happy to provide you with a complimentary replacement antibody, refund or credit per our product warranty.

When should the TransCruz reagent sc-33641 X be used instead of the standard form sc-33641?

Asked by: jenniferc
Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.
