PTGIR encodes the human prostacyclin (IP) receptor, a G protein–coupled receptor that binds prostacyclin (PGI2) to regulate cAMP signaling and downstream PKA-dependent transcriptional programs. IP receptor activation modulates vascular tone, platelet responsiveness, and smooth muscle cell behavior, integrating eicosanoid biosynthesis with GPCR second-messenger pathways. PTGIR signaling also interfaces with inflammatory circuits and endothelial homeostasis through regulation of cytokine production and barrier function. Altered prostacyclin–IP receptor axis activity has been associated with cardiopulmonary and vascular dysfunction phenotypes, supporting its relevance in studies of thrombosis, pulmonary vascular biology, and inflammatory disease mechanisms.
IP Receptor CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the PTGIR gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the PTGIR together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the PTGIR open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish IP Receptor protein expression.
This CRISPR knockout system enables efficient generation of PTGIR-deficient cell models for investigation of IP Receptor signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
