Insulin Rα Antibody (MA-20) is a mouse monoclonal IgG2b kappa light chain antibody specifically engineered to target the insulin receptor alpha subunit (Insulin Rα) of human origin in immunoprecipitation (IP) applications. Insulin Rα monoclonal antibody (MA-20) is raised against placental human Insulin Rα, enabling precise detection of the insulin receptor in human samples, and cross-reacts with Insulin Rα from mouse and rat species, providing versatility for studies involving these model organisms. The insulin receptor is a critical heterodimeric protein composed of two extracellular alpha subunits and two transmembrane beta subunits connected by disulfide bonds. The alpha subunits are responsible for insulin binding, which triggers a series of intracellular signaling pathways that regulate glucose uptake, glycogen synthesis, and overall cellular metabolism. Post-translational modifications of Insulin Rα, particularly N-linked glycosylation, are essential for proper protein folding, stability, and cell surface localization. These glycosylation events facilitate the correct assembly and dimerization of the receptor. Additionally, glycosylation influences the receptor′s interaction with other cellular proteins and insulin responsiveness, playing a pivotal role in maintaining metabolic homeostasis. Dysregulation of these post-translational modifications can lead to impaired insulin signaling, contributing to the development of insulin resistance and type 2 diabetes mellitus. Insulin Rα (MA-20) monoclonal antibody is available in both non-conjugated and multiple conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates, allowing for flexible use in diverse experimental setups. Anti-Insulin Rα antibody (MA-20) is recommended for researchers investigating the molecular underpinnings of insulin action and the pathophysiology of diabetes.

To which domain does INSR/Insulin Receptor α Antibody (MA-20): sc-57344 bind to the insulin receptor: the extracellular domain alpha domain (n-terminus)  the intracellular domain or both.

Are there any issues with using sc-57344: insulin Rα (MA-20) mouse monoclonal primary antibody on mouse samples?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent,we recommend using a directly conjugated primary antibody. Rated 5 out of 5 by from Got target bandWe tried the antibody dilution range from 1:300 to 1:800.We can get 125kda target band, The high conceration 1:300 may cause background and non specific bands, I suggested that the concentration is around 1:500, the exposure time should not be too long, we recommend controled in 3min,also I highly recommend do internal control on the same membrance at the same time to test the experiment conditions.
