ILT-2 Antibody (VMP55) is a mouse monoclonal IgG1 kappa light chain antibody that detects ILT-2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and flow cytometry (FCM). ILT-2 Antibody (VMP55) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The immunoglobulin-like transcript 2 (ILT-2), also referred to as CD85 or MIR7, is a 650 amino acid glycoprotein characterized by a 23 amino acid signal peptide, four extracellular C2-type immunoglobulin superfamily domains, and four intracellular immunoreceptor tyrosine-based inhibitory motifs (ITIMs). ILT-2 plays a crucial role in immune regulation by binding to major histocompatibility complex (MHC) class I molecules, which inhibits the activation of natural killer (NK) and T cells, thereby preventing excessive immune responses. The presence of ILT-2 on immune cells, such as monocytes, B cells, and dendritic cells, is vital for maintaining immune homeostasis and tolerance, particularly in preventing autoimmunity and modulating responses to pathogens. Additionally, ILT-2′s interaction with B cell antigen receptor and histocompatibility leukocyte antigens (HLA)-DR can inhibit calcium mobilization in myeloid cells, further underscoring ILT-2′s importance in immune signaling pathways. Upon tyrosine phosphorylation, ILT-2 associates with tyrosine phosphatase SHP-1, which is essential for ILT-2′s inhibitory function, making ILT-2 a significant target for research in immunology and therapeutic interventions.

Does this antibody recognize monkey ILT2?

Thank you for your question. Anti-ILT-2 Antibody (VMP55) is recommended for detection of ILT-2 of mouse, rat and human origin by WB, IP, IF and FCM. It has not been tested in simian species, so we cannot say whether or not it would work in such samples.

Can ILT-2 (VMP55): sc-20065 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, ILT-2 (VMP55): sc-20065 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
