Mouse Il9 encodes interleukin-9 (IL-9), a pleiotropic cytokine predominantly produced by Th9 cells that modulates immune cell proliferation, differentiation, and survival. IL-9 signals through the IL-9 receptor complex to activate JAK/STAT pathways, with prominent STAT5-dependent transcriptional programs that shape cytokine networks and tissue inflammation. In murine systems, IL-9 influences mast cell and eosinophil biology, mucosal barrier responses, and airway immune remodeling, linking it to allergic inflammation and immune dysregulation. Altered IL-9 activity is frequently studied in models of asthma-like airway inflammation, helminth-driven immunity, and tumor-associated immune microenvironments.
IL-9 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Il9 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Il9 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Il9 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish IL-9 protein expression.
