IL27RA encodes IL-27 receptor alpha (IL-27Rα, also known as WSX-1/TCCR), a cytokine receptor subunit that forms a signaling complex with gp130 to mediate responses to IL-27. Engagement of IL-27Rα activates JAK/STAT pathways, prominently STAT1 and STAT3, shaping T cell differentiation, limiting excessive inflammation, and modulating antigen-presenting cell function. IL27RA-dependent signaling influences Th1/Th17 balance, regulatory networks, and interferon-responsive gene programs that intersect with innate and adaptive immune crosstalk. Dysregulation of this axis has been linked to immune-mediated pathology and altered host defense, making IL27RA a useful target for mechanistic studies in inflammatory and infectious disease models.
IL-27Rα CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the IL27RA gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the IL27RA together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the IL27RA open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish IL-27Rα protein expression.
