Il1r2 encodes mouse interleukin-1 receptor type II (IL-1RII), an atypical member of the IL-1 receptor family that functions predominantly as a decoy receptor for IL-1α and IL-1β. By binding IL-1 ligands without productive signaling, IL-1RII limits ligand availability for IL-1R1 and dampens downstream inflammatory cascades that otherwise engage NF-κB and MAPK-dependent transcriptional programs. IL-1RII activity helps tune innate immune activation, leukocyte recruitment, and cytokine network amplification in myeloid and stromal compartments. Altered IL-1RII expression or shedding has been associated with dysregulated inflammatory responses relevant to autoimmune inflammation, infection biology, and tissue injury models.
IL-1RII CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Il1r2 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Il1r2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Il1r2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish IL-1RII protein expression.
This CRISPR knockout system enables efficient generation of Il1r2-deficient cell models for investigation of IL-1RII signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
