IgM Antibody (M11) is a mouse monoclonal IgG1 antibody that detects Immunoglobulin M (IgM) in human samples through western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). IgM is the largest circulating antibody molecule in humans, consisting of a heavy chain (µ-chain) and a light chain (κ- or λ-chain), forming a pentameric structure with ten binding sites, although steric hindrance prevents simultaneous binding at all sites. The unique structure of IgM is crucial for immune response, exhibiting high avidity due to its polymeric nature, making IgM particularly effective at activating the complement system, which is essential for opsonization and lysis of pathogens. IgM is the first immunoglobulin produced by mature B cells during an immune response, appearing early in infection, which underscores IgM′s importance in initial defense against pathogens. While primarily found in serum, the presence of the J chain allows IgM to function as a secretory immunoglobulin, contributing to mucosal immunity. The IgM chain C, which refers to the constant region of the IgM heavy chain, plays a significant role in immune regulation, further highlighting IgM′s importance in maintaining immune homeostasis.
