IgM Antibody (A-7) is a mouse monoclonal IgM antibody that detects Immunoglobulin M (IgM) in human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). IgM is the largest circulating antibody molecule in humans, consisting of a heavy chain (µ-chain) and a light chain (κ- or λ-chain), along with five base units and ten binding sites, although IgM cannot bind all ten simultaneously due to steric hindrance. The structure of IgM is particularly important as IgM typically exists as a pentamer, allowing for high avidity and effective complement activation, which is crucial for the immune response. Additionally, IgM can form hexamers and is characterized by a polymeric nature, which enhances IgM′s ability to bind antigens and activate the complement system, making IgM a key player in the early stages of infection. While IgM is predominantly found in serum, IgM′s presence in the interstitium is minimal due to size, and IgM plays a significant role as a secretory immunoglobulin thanks to the J chain. As the first immunoglobulin expressed by mature B cells, IgM is essential for initiating the immune response, appearing early during infections and serving as a critical marker for immune system activity.
