Hugl-1 Antibody (A-2) is a mouse monoclonal IgG1 antibody that detects Hugl-1 in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Hugl-1 is a crucial cortical cytoskeleton protein that plays a significant role in regulating mitotic spindle orientation, differentiation, proliferation, and organization of neuroepithelial cells. Hugl-1 is primarily localized in the cytoplasm, where Hugl-1 participates in a complex essential for maintaining cell polarity and epithelial integrity. Hugl-1 interacts with several key partners, including nonmuscle Myosin II heavy chain, PRKCI/aPKC, PARD6B/Par-6, PARD6A, and STX4A, which are vital for cellular functions such as adhesion and migration. Notably, Hugl-1 functions as a tumor suppressor in humans, and Hugl-1 expression is critical for preventing progression of colorectal cancer and melanoma. The gene encoding Hugl-1, LLGL1, shares significant homology with the Drosophila tumor suppressor gene l(2)gl, which encodes Lgl protein, underscoring evolutionary conservation of its function in maintaining cellular architecture. Hugl-1 expression is observed in various tissues, including kidney, brain, and muscle, where Hugl-1 enhances cell adhesion and reduces cell migration, highlighting importance in tissue organization and integrity.
