HS3ST3B1 encodes heparan sulfate glucosamine 3-O-sulfotransferase 3B1, a Golgi-localized enzyme that catalyzes 3-O-sulfation of heparan sulfate chains. This modification helps define heparan sulfate binding sites for extracellular ligands and shapes cell-surface and extracellular matrix signaling microenvironments. By modulating heparan sulfate fine structure, HS3ST3B1 can influence processes including growth factor and chemokine signaling, cell–cell interactions, and adhesion-dependent migration. Altered heparan sulfate sulfation patterns are associated with dysregulated signaling networks implicated in cancer biology, inflammation, and tissue remodeling, making HS3ST3B1 a useful target for mechanistic studies of glycosaminoglycan-dependent pathways.
HS3ST3B1 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the HS3ST3B1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the HS3ST3B1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the HS3ST3B1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish HS3ST3B1 protein expression.
