Hrs Antibody (D-6) is a mouse monoclonal IgG2a antibody that detects hepatocyte growth factor-regulated tyrosine kinase substrate (Hrs) in mouse, rat, and human samples through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-Hrs antibody (D-6) recognizes Hrs, a crucial zinc-finger protein that plays a significant role in intracellular trafficking and signal transduction, particularly associated with early endosomes. Hrs (D-6) monoclonal antibody targets a unique structure including a phosphatidylinositol 3-phosphate-binding domain, which is vital for endosomal targeting, allowing Hrs to co-localize with Clathrin at the carboxy-terminus through a Clathrin box motif. This interaction is essential for proper functioning of endosomal sorting processes, which are critical for maintaining cellular homeostasis and regulating signaling pathways. Additionally, Hrs is involved in ventral folding morphogenesis and shares structural similarities with the yeast protein Vps27p, known for its role in vacuolar protein sorting. The human Hrs gene, located on chromosome 17q25, encodes a 777 amino acid protein, and in Schwann cells, Hrs co-localizes with the tumor suppressor protein schwannomin at endosomes, indicating a potential collaborative role in Hrs-mediated cell signaling.
