HPV16 E1/E4 Antibody (TVG 402) is a mouse monoclonal IgG1 kappa light chain antibody that detects the HPV16 E1/E4 protein of HPV origin by western blotting (WB), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). HPV16 E1/E4 (TVG 402) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The human papillomavirus (HPV) family includes HPV16, a high-risk sexually transmitted virus associated with several cancers, including cervical, anal, vulvar, head, neck, and penile cancers. HPV16 E1/E4 protein plays a crucial role in modulating the cell cycle by inducing G2 phase arrest, which is essential for viral replication. HPV16 E1/E4 achieves this function by interacting with cyclin-dependent kinase 1 (Cdk1) and cyclin B1 complexes, preventing their nuclear entry and thereby inhibiting mitotic progression. Additionally, HPV16 E1/E4 interacts with cyclin A and Cdk2 during the G2 phase, enhancing its ability to block cell division. Notably, HPV16 E1/E4 also associates with keratin intermediate filaments, leading to the disruption of the keratin network, which is significant for the viral life cycle and pathogenesis. The ability of HPV16 E1/E4 to manipulate cellular processes underscores its importance in HPV-related oncogenesis and makes anti-HPV16 E1/E4 antibody (TVG 402) a valuable tool for research into HPV biology and its associated diseases.

Can HPV16 E1/E4 (TVG 402): sc-53324 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Asked by: Professor Griffin
Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, HPV16 E1/E4 (TVG 402): sc-53324 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
