HoxD3 Antibody (4AY) is a mouse monoclonal IgG2a kappa light chain antibody that detects HoxD3 protein of mouse, rat, and human origin by western blotting (WB) and immunoprecipitation (IP). Anti-HoxD3 antibody (4AY) is available as a non-conjugated format. HoxD3, a member of the homeobox gene family, plays a crucial role in regulating gene expression during embryonic development, particularly in establishing the anterior-posterior body axis. HoxD3′s ability to bind DNA as a transcription factor is significantly enhanced through dimerization with PBX homeoproteins, which increases affinity for target genes. This interaction is vital for proper regulation of genes involved in developmental processes. HoxD3 is essential for normal development and plays a pivotal role in wound healing by promoting angiogenesis and collagen synthesis, making HoxD3 a potential therapeutic target for improving healing in diabetic patients with chronic wounds. Research has shown HoxD3 overexpression in several melanoma cell lines, suggesting involvement in cancer progression. HoxD3 enhances motility and invasiveness of cancer cells by upregulating Integrin β3 expression, which may contribute to invasive and metastatic potential of tumors through both TGFβ-dependent and independent pathways.

Using HOXD3 (4AY) antibody for IF. How much antibody would you recommend?

Thank you for your question. We have not yet tested this antibody for immunofluorescent staining and so IF is not a recommended application at this time. If you wish to test the antibody for IF, we would typically recommend a starting dilution of 1:50 (range of 1:25-1:500) for IF.

I am using HoxD3 (4AY): sc-130378 antibody for immunoprecipitation, how much antibody should I use?

Thank you for your question. We recommend using 1–10 µl (i.e., 0.2–2 µg) of antibody with 1 ml of cell lysate. However, optimal antibody concentration should be determined by titration.
