HM74 Antibody (D-8) is a mouse monoclonal IgG1 kappa light chain antibody that detects HM74 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). HM74 (D-8) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. HM74, also known as PUMAG or Puma-g, is a member of the G protein-coupled receptor (GPCR) superfamily, which plays a crucial role in cellular signaling. Specifically, HM74 is a G(i) protein-coupled receptor that mediates nicotinic acid′s metabolic effects, making HM74 significant in regulating lipid metabolism and energy homeostasis. This receptor is predominantly found in the cell membrane and is highly expressed in adipocytes, immune cells, and the spleen, highlighting HM74′s importance in metabolic processes and immune responses. Notably, HM74 is encoded by two different genes in humans (GPR109A and GPR109B), resulting in two homologous proteins, HM74A and HM74B, which share 96% sequence similarity. In contrast, mice and rats possess a single gene (Gpr109a) that encodes HM74 . HM74′s unique structural features, including seven transmembrane domains and the absence of N-linked glycosylation sites near the N-terminus, suggest distinct ligand specificity compared to other GPCR family members, further emphasizing HM74′s specialized role in metabolic regulation.

I am using HM74 (D-8): sc-377292 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?
