His-Tag Antibody (H-3) is a mouse monoclonal IgG1 kappa light chain antibody raised against a His-tagged recombinant protein, designed for applications such as western blotting (WB) and immunoprecipitation (IP). The His-tag consists of a sequence of six histidine residues commonly fused to recombinant proteins to facilitate purification and detection. His-tagged proteins bind strongly to metal ions like nickel or cobalt, allowing efficient purification through immobilized metal affinity chromatography. While His-tags are not typically subject to post-translational modifications, fusion of His-tags to proteins can influence or be influenced by post-translational modifications of the target protein, such as phosphorylation, glycosylation, or ubiquitination. These modifications are crucial because they can affect protein function, localization, stability, and interactions within the cell. Understanding and detecting these changes is important for studying protein behavior in various biological processes and disease states. anti-His-Tag antibody (H-3) enables researchers to accurately detect and analyze His-tagged proteins, even when post-translational modifications are present, thus advancing research in protein expression, function, and therapeutic development.

Thank you for your question. If you are referring to blocking we recommend 5%milk in TBST. The recommended starting dilution range is 1:100-1:1000.

Does this antibody work against RGS.6His tag?

Thank you for your question. Anti-His-Tag Antibody (H-3): sc-8036 can be used to detect any recombinant protein containing a conventional His tag and would be suitable for this application as well.

What buffer should i use to dilute the antibody? can the antibody be stored in that buffer?

Thank you for your question. This depends on the application you are using the antibody for, so it is difficult to answer. For Western blotting we recommend 5% milk with TBS-T, and for IHC we would recommend using a blocking serum or reagent.
