HB-EGF Antibody (G-11) is a mouse monoclonal IgG2a kappa light chain antibody designed to detect Heparin-binding EGF-like growth factor (HB-EGF), also known as HBEGF or the 87-amino acid EGF-like glycoprotein. HB-EGF monoclonal antibody (G-11) targets amino acids 121-208 of the mature human HB-EGF protein, ensuring precise binding and reliable detection in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA) assays. HB-EGF is a pivotal member of the epidermal growth factor (EGF) family, functioning as a potent mitogen by binding to the EGF receptor (EGFR) and heparin sulfate proteoglycans on the cell surface, which is essential for regulating cell proliferation, differentiation, and survival. The HB-EGF gene encodes a 208 amino acid precursor that undergoes enzymatic cleavage to produce the active, soluble form of 86 amino acids, highlighting its critical role in cellular signaling processes. Additionally, the membrane-bound form of HB-EGF serves as the diphtheria toxin receptor, underscoring its importance in both physiological and pathological contexts. I can't get the antibody to work on WB of cell lysate from Human and CHO cells-lines. I followed the protocols as cited unfortunately it is not working. Any recommendations, please?

Could this HB-EGF Antibody (G-11): sc-74441 be used to detect cytoplasmic tail fragment form of HB-EGF?

Could this HB-EGF Antibody (G-11): sc-74441 be used for mouse cells western blot? Could this antibody detect both precursor and mature form of HB-EGF, eliciting multiple target bands for mouse cells western blot?

Thank you for your question. This HB-EGF antibody was made against the mature form, but can detect both the precursor and mature form of HB-EGF. It is only recommended for human samples, however we do have a citation using it on mouse samples (PMID: 24244689). Can HB-EGF (G-11): sc-74441 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, HB-EGF (G-11): sc-74441 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
