GPR171 encodes an orphan class A (rhodopsin-like) G protein-coupled receptor that is predominantly coupled to inhibitory signaling and is implicated in modulating intracellular cAMP and downstream transcriptional programs. It is expressed in immune and neural-associated contexts, where it has been linked to regulation of cell signaling tone, neuromodulatory circuits, and inflammatory responsiveness. Reported interactions with the neuropeptide BigLEN suggest a role in peptide-mediated GPCR signaling and network-level control of cellular activation states. Altered GPCR signaling involving GPR171 has been explored in relation to neurobehavioral phenotypes and immune regulation, supporting its relevance for pathway-level studies of receptor-mediated signal transduction.
GPR171 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the GPR171 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the GPR171 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the GPR171 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish GPR171 protein expression.
This CRISPR knockout system enables efficient generation of GPR171-deficient cell models for investigation of GPR171 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
