SEE ALSO...

GnRH II Antibody (D-9) is a mouse monoclonal IgG1 kappa light chain antibody that detects GnRH II protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-GnRH II antibody (D-9) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. GnRH II plays a crucial role in reproductive functions, particularly in the midbrain, where GnRH II is involved in modulating gonadotropin release. This decapeptide is essential for the hypothalamic-pituitary-gonadal axis and exhibits significant expression in tissues outside the brain, such as kidney, bone marrow, and prostate, indicating potential involvement in various physiological processes beyond reproduction. GnRH II′s unique expression pattern suggests interactions with specific proteins in these tissues, influencing cellular signaling pathways and contributing to functions such as cell proliferation and differentiation. Through binding to specific G protein-coupled receptors in the pituitary, GnRH II regulates luteinizing hormone and follicle-stimulating hormone synthesis and secretion, maintaining a vital role in reproductive health and development.

Can GnRH II (D-9): sc-25344 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: Cweed
Thank you for your question. Yes, GnRH II (D-9): sc-25344 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining

Rated 4 out of 5 by from Primate retinaAntibody was tested under standard conditions (Long Y, Bordt AS, Liu WS, Davis EP, Lee SJ, Tseng L, Chuang AZ, Whitaker CM, Massey SC, Sherman MB, Marshak DW. Wide-field diffuse amacrine cells in the monkey retina contain immunoreactive Cocaine- and Amphetamine-Regulated Transcript (CART). Peptides. 2016 Oct;84:22-35). The isolated retina of a baboon was fixed for 60 minutes in 4% paraformaldehyde. Tissue was incubated for 9 days in a 1:1,000 solution of primary antibody with detergent and a blocking agent at 4 °C, washed, then incubated in an affinity-purified secondary antibody for 4 days at 4 °C, washed, and mounted in medium containing DAPI. No peptide blocking control was performed. Results: Light punctate labeling and low background throughout the optic fiber layer, ganglion cell layer, inner plexiform layer, and inner nuclear layer (less).
