Glypican-3 Antibody (F-3) is a mouse monoclonal IgM antibody that detects glypican-3 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-glypican-3 antibody (F-3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Glypican-3 (GPC3) is an integral membrane protein that plays a crucial role in regulating cell growth and development, particularly during embryogenesis. GPC3 is a member of the glypican family of heparan sulfate proteoglycans, which are attached to the cell membrane via a glycosyl-phosphatidyl-inositol (GPI) anchor. GPC3 is primarily expressed in embryonic mesodermal tissues such as the lung, liver, and kidney, where GPC3 interacts with growth factors like insulin-like growth factor II (IGF-II) and fibroblast growth factor 2 (FGF-2) to modulate cellular signaling pathways that influence tissue and organ growth. Mutations in the GPC3 gene are associated with Simpson-Golabi-Behmel syndrome (SGBS), a recessive X-linked condition characterized by pre- and post-natal overgrowth, highlighting GPC3′s significance in developmental processes. GPC3 expression can be downregulated through mechanisms such as promoter hypermethylation or the repression of specific transcription factors, which may have implications in various pathologies, including cancer.

What secondary would you recommend for western blot since it is an IgM monoclonal? Most secondary's are to IgG and I not 100% sure they will cross-react to IgM.

What application is the blocking peptide sc-390587 P appropriate for?

Asked by: Professor Griffin
Thank you for your question. The blocking peptide is intended for use as a negative control, by pre-adsorbing the mouse monoclonal antibody against the antigen. For full protocol details, please contact our Technical Services Department or view our online protocol here: https://www.scbt.com/scbt/resources/protocols/peptide-neutralization
