Gfi-1 Antibody (G-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects Gfi-1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-Gfi-1 antibody (G-11) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Growth factor independent 1 (Gfi-1) plays a crucial role as a transcriptional repressor, specifically binding to the DNA consensus sequence TAAATCAC(A/T)GCA, which is vital for regulating gene expression involved in cell survival and differentiation. Gfi-1′s structure includes six C2H2-type zinc finger motifs located at the carboxy-terminus, with zinc fingers 3, 4, and 5 being essential for DNA binding capability. A 20 amino acid SNAG domain in Gfi-1 is instrumental in mediating transcriptional repression, particularly of the pro-apoptotic gene Bax, thereby inhibiting cell death. Gfi-1 expression occurs in the lymphoid system, granulocytes, and activated macrophages, highlighting Gfi-1′s importance in immune responses. Gfi-1B, a closely related protein, is primarily expressed in the bone marrow and spleen and serves as a direct repressor of the p21 promoter as well as the Socs 1 and 3 promoters, indicating a broader regulatory role in hematopoiesis and immune function. Human Gfi-1 and Gfi-1B genes are located on chromosomes 1p22 and 9q34.3, respectively, underscoring their significance in human health and disease.
