Gfi-1 Antibody (B-9) is a mouse monoclonal IgG1 kappa light chain antibody that detects Gfi-1 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-Gfi-1 antibody (B-9) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Growth Factor Independent 1 (Gfi-1) plays a crucial role as a transcriptional repressor, specifically binding to the DNA consensus sequence TAAATCAC(A/T)GCA, which is vital for regulating gene expression involved in cell survival and differentiation. Gfi-1′s structure includes six C2H2-type zinc finger motifs located at the carboxy-terminus, with zinc fingers 3, 4, and 5 being essential for DNA binding capability. Gfi-1 contains a 20 amino acid SNAG domain that is critical for transcriptional repression function, particularly in inhibiting the pro-apoptotic gene Bax at both the mRNA and protein levels, thereby preventing cell death. Gfi-1 is predominantly expressed in granulocytes and activated macrophages, extending influence beyond the lymphoid system, while Gfi-1B is primarily found in the bone marrow and spleen, where Gfi-1B also functions as a transcriptional repressor. The genes encoding human Gfi-1 and Gfi-1B are located on chromosomes 1p22 and 9q34.3, respectively, highlighting their genetic significance in cellular processes.
