GDF-15 Antibody (H-2) is a mouse monoclonal IgG2a kappa light chain antibody that detects growth differentiation factor 15 (GDF-15) in mouse, rat, and human samples through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-GDF-15 antibody (H-2) is available in non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. GDF-15, also known as PDF, MIC-1, PLAB, NAG-1, and PTGF-β, belongs to the transforming growth factor beta (TGF-beta) superfamily and plays a crucial role in cellular responses to stress and inflammation. GDF-15 is synthesized intracellularly and secreted as a dimer linked by disulfide bonds, with highest expression in epithelial cells and macrophages, while maintaining wide distribution in adult tissues. GDF-15 is secreted into the cerebrospinal fluid from the choroid plexus in the brain, highlighting importance in central nervous system function. p53 tumor suppressor protein regulates GDF-15 gene expression, and in cultured cerebellar granule neurons, GDF-15 prevents cell death by activating the Akt pathway while inhibiting ERK signaling. This trophic factor promotes cell survival and differentiation in specific neuronal populations. GDF-15 overexpression in prostate cancer may serve as a potential diagnostic marker. During pregnancy, GDF-15 may help suppress proinflammatory cytokines derived from the mother, enhancing fetal survival.

I have a question. I did WB to detect gdf15 in H9C2 cell by use the GDF-15 antibody (H-2): sc-515675. I got 2 band : one above 50 kDa, the other around 35 kDa. is this the right phenomenon?

I have a question. is western blot data non-reducing conditions or reducing conditions? I did WB to detect gdf15 in LNCaP by non-reducing conditions. I got 2 band : one above 40 kDa, the other around 35 kDa.

Thank you for your question. Our Western blot data with this antibody is under reducing conditions. We have not tested the antibody under non-reducing conditions.

I have a rat cell line, can this ab detect GDF15 from rat?

Thank you for your question. Yes, this antibody is recommended for detection of GDF15 of mouse, rat and human origin. We have Western blot data on our website in the rat cell line, RIN-m5F.

Has anyone any information that this Ab can detect human GDF15. According to the protein alignments in aa 41-62 the Human sequence differs by an additional 7 amino acids. Would be keen to see some experimental data if anyone has it. Thanks
