GCK Antibody (G-5) is a mouse monoclonal IgG1 antibody that detects glucokinase (GCK) in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). GCK plays a crucial role in glucose metabolism, primarily functioning as a glucose sensor in pancreatic beta cells, where GCK regulates insulin secretion in response to glucose levels. This regulation is vital for maintaining glucose homeostasis in the body, as GCK determines the threshold for insulin release, thereby influencing blood sugar levels. Additionally, GCK is also expressed in the liver, where GCK catalyzes the phosphorylation of glucose, marking the first committed step in glucose disposal. The activity of GCK is essential; any deficiency can lead to impaired insulin secretion and hyperglycemia, conditions that are often associated with maturity onset diabetes of the youth (MODY). Notably, mutations in the GCK gene have been linked to this form of diabetes, highlighting GCK′s significance in metabolic health. Anti-GCK antibody (G-5) is an invaluable tool for researchers studying glucose metabolism and its implications in diabetes and other metabolic disorders.

What blocking solution should I use with GCK (G-5): sc-55496 monoclonal antibody for Western blot?

Asked by: Cweed
Thank you for your question. We recommend blocking with Blotto (either TBS Blotto A: sc-2333 or TBS Blotto B: sc-2335). Our Western blot protocol can be found here: https://www.scbt.com/scbt/resources/protocols/western-immuno-blotting
