Gαi-3 Antibody (H-7) is a mouse monoclonal IgG3 kappa light chain antibody engineered to recognize Gαi-3, also known as GNAI3, from rat origin. Anti-Gαi-3 antibody (H-7) targets an epitope spanning amino acids 339-354 at the C-terminus of Gαi-3, enabling detection in mouse, rat, human, and additional species including equine, canine, bovine, porcine, and avian samples through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA) assays. Gαi-3 is a member of the heterotrimeric G protein family, which plays a crucial role in transducing signals from cell surface receptors to intracellular effectors. Structurally, Gαi-3 consists of a GTP-binding domain and a helical domain, essential for binding and hydrolyzing GTP, which regulates activation and interaction with downstream effectors such as adenylyl cyclase. Post-translational modifications, including myristoylation and phosphorylation, are vital for proper localization and function of Gαi-3, influencing interaction with receptors and other signaling molecules. Gαi-3 interacts with various proteins, including G protein-coupled receptors (GPCRs), beta-gamma subunits, and effectors like ion channels and kinases, facilitating diverse cellular responses such as inhibition of adenylyl cyclase, modulation of ion channel activity, and regulation of cell growth and differentiation. Gαi-3 (H-7) monoclonal antibody is available in both conjugated and non-conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing versatile options for experimental applications. Does this antibody work well in Raw 264.7 and Jurkat cell lines?

How likely is it that this antibody will show a cross reaction to Gnai2 since they are similar in structure? Was this tested? How similar is the immunogen the antibody was raised against to Gnai2? Thanks in advance!

Thank you for your question. This mouse monoclonal antibody, Gα i-3 (H-7): sc-365422, is specific for an epitope mapping between amino acids 339-354 at the C-terminus of Gα i-3 of rat origin. This region does share significant identity (80%) with the Gα i-2 protein, so there is potential of cross-reactivity. Unfortunately, this cross-reactivity has not yet been tested.
