Galectin-3 Antibody (M3/38) is a rat monoclonal IgG2a antibody engineered to target galectin-3, also known by its alternate names Mac-2, hMac-2 (human Mac-2), GALBP (galactose-binding protein), CBP35 (carbohydrate-binding protein 35), and LGALS3 (lectin, galactoside-binding, soluble, 3). Anti-galectin-3 antibody (M3/38) is raised against galectin-3 of mouse origin and is recommended for the detection of galectin-3 in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). Galectin-3 is a pivotal member of the galectin family of soluble β-galactoside-binding lectins that play essential roles in modulating cell-to-cell adhesion and interactions between cells and the extracellular matrix (ECM), which are critical for processes such as tissue development, immune response, and cellular communication. Located both intracellularly in the cytoplasm and nucleus, and extracellularly in the ECM, galectin-3′s cellular localization is crucial for its diverse functions. Intracellularly, galectin-3 is involved in signaling pathways, pre-mRNA splicing, and apoptosis, while extracellularly, galectin-3 facilitates cell adhesion, migration, and interactions with the ECM. These functions make galectin-3 integral to maintaining cellular homeostasis and responding to cellular stress. Importantly, galectin-3 is implicated in tumor progression and metastasis by influencing cancer cell proliferation, apoptosis resistance, and angiogenesis, thereby making galectin-3 a significant target in cancer research. Galectin-3 (M3/38) antibody is available in both non-conjugated form and multiple conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates, providing versatile options for diverse experimental applications.

Que dilución es recomendable usar para WB en suero humano

Asked by: Mari30
Thank you for your question. Galectin-3 Antibody (M3/38): sc-23938 is recommended for detection of galectin-3 of mouse, rat and human origin by Western Blotting at a starting dilution 1:200 (general dilution range 1:100-1:1000)

I would like to stain galectin-3 on renal sections by IHC. Which demasking conditions do you recommend for this antibody (EDTA, citrate, etc.) ? Which dilution as well ?

Asked by: pierrelo
Thank you for your question. Our recommended method is as follows: Heat treatment: Place slides in a container and cover with 10 mM sodium citrate buffer, pH 6.0 (sc-294091); or with 50 mM glycine-HCl buffer (sc-29096), pH 3.5, with 0.01% (w/v) EDTA (sc-29092). Heat at 95° C for 5 minutes. Top off with fresh buffer and heat at 95° C for 5 minutes (optimal incubation time may vary for each tissue type). Allow slides to cool in the buffer for approximately 20 minutes. Wash in deionized H2O three times for 2 minutes each. Aspirate excess liquid from slides.
