FUT2 CRISPR/Cas9 KO Plasmid (h) | sc-409559 | 20 µg | $397.00 |
FUT2 encodes α(1,2)-fucosyltransferase 2, a Golgi-resident glycosyltransferase that catalyzes terminal fucosylation of glycans to generate H type 1/2 antigens on mucosal epithelial surfaces and in secretions. By controlling fucosylated glycan motifs on glycoproteins and glycolipids, FUT2 influences protein trafficking, epithelial barrier properties, and host–microbe interactions at the gut and airway mucosa. Genetic variation and altered FUT2 activity are linked to secretor status and associated differences in mucosal glycosylation patterns that correlate with susceptibility to infectious and inflammatory phenotypes. As a key regulator of glycan-dependent recognition events, FUT2 is widely studied in glycomics, microbiome biology, and epithelial immunology.
FUT2 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the FUT2 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the FUT2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the FUT2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish FUT2 protein expression.
This CRISPR knockout system enables efficient generation of FUT2-deficient cell models for investigation of FUT2 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
