FR Antibody (E-11) is a mouse monoclonal IgG2a kappa light chain antibody that detects folate receptor (FR) in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). FR Antibody (E-11) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing versatility for different experimental needs. The folate receptor family plays a crucial role in cellular uptake of folate, an essential vitamin necessary for DNA synthesis and repair, as well as amino acid metabolism. This family consists of five members: α, β, γ, γ′ (produced by alternative splicing), and δ, with α-FR and β-FR being particularly significant as they are expressed in malignant tissues, making them valuable targets for cancer therapy. The unique structure of these receptors, which includes a glycosylphosphatidylinositol (GPI) anchor for α-FR and β-FR, facilitates their localization to the cell membrane, enhancing their ability to mediate the endocytosis of 5-methyltetrahydrofolate. This mechanism is vital for maintaining folate homeostasis in cells, especially in rapidly dividing tissues, and underscores the importance of folate receptors in both normal physiology and cancer biology. α-FR serves as a highly selective tumor marker, offering potential for targeted drug delivery systems that utilize folate derivatives to selectively transport therapeutic agents to tumor cells, thereby improving treatment efficacy while minimizing systemic toxicity.

Thank you for your question. FR Antibody (E-11) is only recommended for detection of α-FR, β-FR and γ-FR of human origin and α-FR, and β-FR of mouse and rat origin by WB, IP, IF, IHC(P) and ELISA. The antibody is also available conjugated to Alexa Fluor® 488, Alexa Fluor® 546, Alexa Fluor® 594 or Alexa Fluor® 647 for WB (RGB), IF, IHC(P) and FCM.

Can I use this product to detect murine macrophages (raw 264.7) FR using goat anti-mouse secondary antibody?

Thank you for your question. This FR antibody is recommended for detection in mouse samples. We have not specifically tested the antibody in RAW 264.7. This is a mouse monoclonal IgG2a, and so it can be detected using a secondary antibody that is goat anti-mouse IgG.
