FOSB encodes Fos B, an immediate early transcription factor of the AP-1 complex that dimerizes with JUN family proteins to regulate stimulus-dependent gene expression. Fos B integrates signals from MAPK/ERK, calcium, and cAMP pathways to control programs linked to cell proliferation, differentiation, survival, and synaptic plasticity. Its rapid induction following growth factor exposure, stress, and neuronal activity makes it a useful marker and mediator of activity-dependent transcription. Dysregulated FOSB/AP-1 signaling has been associated with oncogenic transformation and invasion in multiple tissue contexts, as well as neuropsychiatric and neurodegeneration-related transcriptional remodeling.
Fos B CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the FOSB gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the FOSB together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the FOSB open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Fos B protein expression.
This CRISPR knockout system enables efficient generation of FOSB-deficient cell models for investigation of Fos B signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
